ALY117 is a bispecific T-cell engager (TCE) being developed by ATLyphe, a spin-off from ETH Zurich and the University of Zurich, for use as a precision conditioning agent prior to hematopoietic stem cell transplantation (HSCT). The antibody-based molecule is designed to target CD117 (c-Kit), a receptor highly expressed on hematopoietic stem cells (HSCs) and leukemic blasts, while simultaneously binding to CD3 on T cells. This dual binding recruits and activates the patient's own T cells to selectively eliminate CD117-positive cells, including both healthy stem cells and residual malignant cells in conditions like Acute Myeloid Leukemia (AML) and Myelodysplastic Syndrome (MDS). By replacing or reducing the need for intensive chemotherapy and total body irradiation, ALY117 aims to minimize the severe toxicities associated with traditional conditioning regimens and reduce the risk of post-transplant relapse by clearing minimal residual disease.
