AM-1638 is a potent and orally bioavailable small molecule that acts as a full agonist of the G protein-coupled receptor 40 (GPR40), also known as free fatty acid receptor 1 (FFA1). It was initially developed by Amgen. The drug has demonstrated antidiabetic activity in preclinical models and is being studied for its potential to treat metabolic disorders such as type II diabetes. Mechanistically, activation of GPR40 by AM-1638 enhances insulin secretion in response to elevated glucose levels and exerts protective effects against oxidative stress in various cell types via pathways involving adenosine monophosphate-activated protein kinase (AMPK) and nuclear factor erythroid 2-related factor 2 (NRF2). In animal studies, AM-1638 showed excellent oral bioavailability across species and improved glycemic control. Its development status remains at the preclinical stage[1][2][3][4][5][6][7].
