Overview

**AM-461** is an oral small molecule antagonist of the DP2 receptor (also known as CRTH2 or prostaglandin D2 receptor 2), developed by Amira Pharmaceuticals for inflammatory and respiratory diseases such as asthma. It acts by selectively blocking DP2-mediated signaling, which is implicated in Th2-driven allergic inflammation, type 2 immunity, and conditions like asthma and chronic obstructive pulmonary disease (COPD). As a backup to Amira's lead DP2 antagonist AM211, AM-461 advanced to Phase 1 clinical trials, demonstrating positive pharmacokinetics and safety, before development was transferred following Amira's acquisition by Bristol Myers Squibb.[1][2][3][5][7][8][9]
