AM003 is a first-in-class, individualized immunotherapy developed for the treatment of advanced and metastatic solid tumors. It is a bispecific personalized aptamer composed of two functional arms—a T cell engager arm and a tumor-targeting arm—incorporating cytosine-phosphate-guanine (CpG) nucleotides. The drug acts by targeting toll-like receptor 9 (TLR9), stimulating antigen-presenting cells such as dendritic cells and B cells to enhance immune activation within the tumor microenvironment. This results in increased infiltration and activation of CD4+ and CD8+ T cells as well as B cells, directly inducing tumor cell death while promoting an anti-tumor immune response. Preclinical studies have shown that AM003 can synergize with anti-PD1 therapy to further boost immune activity. The drug is administered intratumorally or locally via subcutaneous injection. Aummune Therapeutics has completed a Phase 1 open-label dose-escalation trial in patients with various solid tumors—including pancreatic, ovarian, lung, head & neck cancers, sarcomas—demonstrating favorable safety and tolerability profiles without dose-limiting toxicities[1][3][4][5][7].
