Overview

AM260 is a small molecule inhibitor of the 5-lipoxygenase activating protein (FLAP), originally developed by Amira Pharmaceuticals. FLAP is a critical protein in the leukotriene synthesis pathway, facilitating the transfer of arachidonic acid to 5-lipoxygenase (5-LO). By inhibiting FLAP, AM260 prevents the production of pro-inflammatory leukotrienes (such as LTB4 and cysteinyl leukotrienes), which are key mediators in the pathogenesis of asthma, chronic obstructive pulmonary disease (COPD), and other inflammatory or fibrotic conditions. Following the acquisition of Amira Pharmaceuticals by Bristol Myers Squibb in 2011, the development of FLAP inhibitors in their portfolio was further explored for indications including idiopathic pulmonary fibrosis (IPF). AM260 demonstrated safety and tolerability in early-stage clinical trials, but its development appears to have been discontinued in favor of other candidates within the BMS pipeline.
