Overview

AM4113 is an orally available small molecule that acts as a neutral antagonist at the cannabinoid CB1 receptor. Developed by MAK Scientific Management, it is primarily being investigated for the treatment of cannabis addiction and other substance use disorders, including nicotine dependence and obesity. Unlike first-generation CB1 inverse agonists such as rimonabant, which were associated with significant psychiatric side effects like depression and anxiety, AM4113 is a neutral antagonist. This means it blocks the receptor's activation by endocannabinoids or exogenous cannabinoids without suppressing its constitutive (baseline) activity, a distinction hypothesized to maintain therapeutic efficacy in reducing drug-seeking behavior while minimizing adverse neuropsychiatric effects. The compound has demonstrated the ability to reduce the reinforcing effects of cannabinoids and withdrawal symptoms in preclinical models.
