Overview

AM6545 is a peripherally selective, highly potent and selective cannabinoid receptor type 1 (CB1) antagonist that does not significantly cross the blood-brain barrier. It was developed to inhibit endocannabinoid signaling by competitively antagonizing CB1 receptors in peripheral tissues. Unlike earlier CB1 antagonists such as rimonabant—which were associated with centrally mediated side effects like depression and nausea—AM6545 acts outside the central nervous system, thereby reducing appetite and improving metabolic parameters without these adverse effects. Preclinical studies have shown that AM6545 can decrease body weight, reduce fat mass, improve dyslipidemia and insulin resistance, and modulate adipokine levels in animal models of obesity. The drug was originally developed by MAKScientific for potential use in treating obesity and metabolic disorders[2][3][4][7].
