AMA006

AMA006 is a locally-acting Rho kinase (ROCK) inhibitor developed by Amakem Therapeutics for the treatment of glaucoma and ocular hypertension. Designed as a "soft drug," AMA006 is engineered to be potent at the site of administration in the eye while undergoing rapid metabolic inactivation upon entering the systemic circulation. This design aims to minimize systemic side effects and local adverse events such as conjunctival hyperemia, which are common with other ROCK inhibitors. By inhibiting ROCK1 and ROCK2, the drug promotes the relaxation of the trabecular meshwork, thereby increasing the outflow of aqueous humor and effectively reducing intraocular pressure (IOP). The compound reached Phase 2 clinical development, demonstrating significant IOP-lowering efficacy and a favorable safety profile.
