Drug intelligence / Profile preview
AMA1-RON2L SBD1 immunogen (Structure-Based Design 1) is an experimental, single-component recombinant protein malaria vaccine candidate designed to elicit strain-transcending protective immunity against the blood-stage of Plasmodium falciparum. Developed by researchers at the National Institute of Allergy and Infectious Diseases (NIAID), SBD1 is a structurally engineered fusion protein that mimics the naturally occurring complex between apical membrane antigen 1 (AMA1) and rhoptry neck protein 2 (RON2L), which is essential for merozoite invasion of host erythrocytes. Unlike traditional AMA1 vaccines that suffer from limited efficacy due to high genetic polymorphism, SBD1 directs neutralizing antibody responses to conserved, strain-transcending epitopes on AMA1 that are independent of the RON2L binding site. In preclinical studies, SBD1 has demonstrated improved thermostability, high production yields, and potent neutralizing activity against diverse strains of Plasmodium falciparum.
