Overview
Amantadine is a synthetic adamantane derivative that acts as a non-competitive N-methyl-D-aspartate (NMDA) receptor antagonist and a weak dopaminergic agonist. Originally developed as an antiviral agent for Influenza A, it is now primarily used to treat Parkinson's disease and drug-induced extrapyramidal symptoms. Amantadine increases dopamine release and inhibits its reuptake, while its NMDA antagonism helps manage levodopa-induced dyskinesia. Beyond its established roles, amantadine has been investigated for treating Multiple Sclerosis-related fatigue and walking impairment, attention-deficit/hyperactivity disorder (ADHD), and aggressive behavior in pediatric populations. It has also shown potential as an antiviral and antidepressant treatment for patients infected with Borna disease virus 1 (BDV-1).
