Overview

Amantadine + imipramine is a combination of two small molecule drugs—amantadine, an adamantane derivative with dopaminergic and NMDA receptor antagonist properties, and imipramine, a tricyclic antidepressant that inhibits the reuptake of norepinephrine and serotonin. This combination has been studied for its synergistic antidepressant-like effects in preclinical models. Research indicates that co-administration produces stronger antidepressant effects than either drug alone in behavioral tests such as the forced swimming test. Mechanistically, repeated joint administration upregulates dopamine D2 and D3 receptors in the brain, which may underlie their enhanced efficacy on depressive symptoms resistant to standard treatments[1][2][3][4]. Amantadine is also known for its antiviral activity (against influenza A) and use in Parkinson's disease; imipramine is primarily used for depression.
