Overview

AMAT CAR T cells are an experimental chimeric antigen receptor (CAR) T-cell therapy designed to target **Mesothelin (MSLN)**, a glycoprotein overexpressed in various solid tumors including mesothelioma, lung, and ovarian cancers. The CAR construct utilizes a high-affinity single-chain variable fragment (scFv) derived from the humanized monoclonal antibody **amatuximab** (also known as MORAb-009). This specific CAR T-cell product incorporates **CD28** as a costimulatory domain and **CD3ζ** as the primary signaling domain. In preclinical research, AMAT CAR T cells serve as a benchmark for high-affinity targeting; notably, while they exhibit subnanomolar affinity for human MSLN, they do not cross-react with mouse MSLN, which limits their utility in assessing on-target, off-tumor toxicities in standard murine models compared to cross-reactive VHH-based constructs.
