Overview

Amatuximab is a chimeric IgG1/kappa monoclonal antibody that targets mesothelin, a cell surface glycoprotein highly expressed in several cancers including malignant pleural mesothelioma, ovarian cancer, and pancreatic adenocarcinoma. By binding to mesothelin on tumor cells, amatuximab triggers an immune-mediated response—primarily through antibody-dependent cellular cytotoxicity (ADCC)—leading to tumor cell lysis. It was developed for use as an antineoplastic and immunotherapeutic agent. Amatuximab was originally developed by Morphotek and later by Eisai. The drug has received orphan drug status for malignant pleural mesothelioma and pancreatic cancer but is not commercially available; its highest development phase was Phase II before discontinuation in these indications[1][2][3][5][6].
