Overview

Amb a 1 Immunostimulatory Conjugate (AIC) is an investigational immunotherapeutic developed by Johns Hopkins University in collaboration with Dynavax Technologies for the treatment of seasonal allergic rhinitis caused by ragweed allergy. The drug consists of the purified major ragweed allergen, Amb a 1, covalently linked to immunostimulatory phosphorothioate oligodeoxynucleotides (ISS-ODN) containing unmethylated CpG motifs. AIC functions by targeting the immune system's Toll-like receptor 9 (TLR9), which promotes a Th1-biased immune response and suppresses the Th2-mediated allergic response typical of ragweed allergy. This shift in the immune profile leads to reduced nasal inflammation, decreased eosinophilia, and improved clinical symptoms during ragweed season. It is typically administered via a short course of subcutaneous injections prior to the onset of the allergy season.
