Amb a 1-immunostimulatory oligodeoxyribonucleotide conjugate (AIC) is an investigational immunotherapeutic compound developed for the treatment of seasonal allergic rhinitis due to short ragweed pollen. It consists of purified Amb a 1 (the major allergen from short ragweed) covalently linked to an immunostimulatory phosphorothioate oligodeoxyribonucleotide (ISS-ODN), which acts as an adjuvant. The mechanism involves stimulating Toll-like receptor 9 (TLR9), promoting Th1 immune responses and regulatory T cell infiltration while reducing Th2 cytokines and eosinophilia in the nasal mucosa after exposure to the allergen. Clinical studies showed that AIC was safe and well tolerated but did not demonstrate significant clinical benefit over placebo in early trials; development was discontinued after phase II[1][2][4].
