Ambrosia ambrosioides pollen extract is an allergenic biologic used for both the diagnosis and treatment of ragweed-induced hypersensitivity. As a therapeutic agent in allergen immunotherapy (AIT), it is administered in gradually increasing doses to induce immunological tolerance. The mechanism involves a shift in the immune response from a Th2-mediated allergic profile to a Th1 or regulatory T-cell (Treg) profile. This modulation leads to the expansion of Treg populations, increased secretion of anti-inflammatory cytokines such as interleukin-10 (IL-10), and the production of allergen-specific IgG4 antibodies. These IgG4 antibodies act as 'blocking antibodies' that compete with IgE for allergen binding, thereby preventing the degranulation of mast cells and basophils. In diagnostic applications, the extract is used in skin prick testing to identify patients with specific IgE-mediated sensitivity to canyon ragweed.
