Ambuzutinib (AK119) is a humanized monoclonal antibody targeting CD73 (ecto-5'-nucleotidase), an enzyme that plays a critical role in the adenosine pathway within the tumor microenvironment. By binding to and inhibiting the enzymatic activity of CD73, ambuzutinib prevents the conversion of adenosine monophosphate (AMP) into adenosine. Adenosine is a potent immunosuppressive molecule that hinders the activity of T cells and natural killer (NK) cells; therefore, reducing its concentration helps to restore and enhance anti-tumor immune responses. Developed by Akeso, ambuzutinib is primarily being investigated for the treatment of advanced solid tumors, often in combination with checkpoint inhibitors like PD-1 antibodies to overcome immune resistance. It has also been explored in clinical trials for its potential to modulate B-cell immune responses in the context of viral infections such as COVID-19.
