Amcasertib (BBI503) is an orally administered, first-in-class small molecule inhibitor that targets cancer stemness kinases. It acts by inhibiting multiple serine-threonine kinases involved in the maintenance and survival of cancer stem cells, including the suppression of key pathways such as NANOG and CD133. By disrupting these pathways, amcasertib induces apoptosis and inhibits proliferation, invasion, migration, epithelial-mesenchymal transition (EMT), colony formation, and spheroid growth in various cancer cell models. The drug has shown particular promise in preclinical studies for ovarian cancer—especially estrogen receptor-negative subtypes—and breast cancer stem cells. Amcasertib is being investigated for use as a monotherapy or in combination with other anti-cancer agents across a range of solid tumors including hepatocellular carcinoma, ovarian cancer, cholangiocarcinoma, gastrointestinal stromal tumors (GIST), transitional cell carcinoma (TCC), renal cell carcinoma (RCC), head and neck squamous cell carcinoma (HNSCC), salivary gland cancers, and breast cancer[1][3][4][5][6][7][8].
