**Amcasertib + sorafenib** is an investigational oral combination therapy studied primarily in patients with advanced hepatocellular carcinoma (HCC). **Amcasertib** (BBI503) is a small molecule kinase inhibitor that targets cancer stemness pathways by inhibiting multiple cancer stem cell kinases (including stemness kinases such as Nanog, Stat3, and others). **Sorafenib** is a multi-kinase inhibitor that targets RAF kinase, vascular endothelial growth factor receptors (VEGFR-1, -2, -3), and platelet-derived growth factor receptor beta (PDGFR-β), resulting in inhibition of tumor angiogenesis and cell proliferation. The combination was explored in Phase Ib/II studies and was shown to be safe and feasible at full dosing for both agents, with early evidence of anti-tumor activity in patients with advanced liver cancer[1][5].
