Amediplase is a recombinant chimeric protein, classified as a third-generation thrombolytic agent. It is composed of functional domains from both tissue plasminogen activator (t-PA) and pro-urokinase plasminogen activator (scu-PA), specifically the kringle 2 domain of alteplase and the protease domain of urokinase. Developed by Menarini, it is being investigated for the treatment of thrombosis and myocardial infarction. Its mechanism involves activating plasminogen to plasmin, which then degrades fibrin to dissolve blood clots. Studies suggest it may be a more potent thrombolytic agent than Tenecteplase and scu-PA at therapeutic concentrations, with moderate fibrin binding that does not hinder clot penetration.
