Overview
Amelenodor (NX-13) is an orally administered, gut-selective small molecule agonist of the Nucleotide-binding oligomerization domain, Leucine-rich Repeat containing X1 (NLRX1) receptor. NLRX1 is a mitochondria-associated protein that functions as a negative regulator of inflammatory signaling pathways, including NF-κB and Type I interferon. By activating NLRX1, amelenodor modulates mitochondrial metabolism and reduces the production of pro-inflammatory cytokines, thereby promoting mucosal healing and reducing intestinal inflammation. Originally developed by Landos Biopharma, the program was acquired by AbbVie in 2024 and is currently being evaluated in Phase 2 clinical trials for the treatment of moderate to severe ulcerative colitis.
