Amelparib (JPI-289) is a potent, orally active and water-soluble small molecule inhibitor of poly(ADP-ribose) polymerase 1 (PARP-1). It acts by inhibiting PARP-1 activity in the nanomolar range and has demonstrated neuroprotective effects in preclinical models by reducing infarct volume and apoptosis following ischemic stroke. Amelparib is under development primarily for acute ischemic stroke and myocardial infarction. The drug is being developed by Jeil Pharmaceutical and has reached Phase 2 clinical trials for both acute ischemic stroke and myocardial infarction. Its mechanism of action involves inhibition of necrosis, apoptosis, and inflammation through PARP-1 blockade[1][3][4][5][6][7].
