Overview

Amelubant is a potent, orally bioavailable small molecule that acts as a competitive antagonist of the leukotriene B4 (LTB4) receptors, specifically BLT1 and BLT2. Developed by Boehringer Ingelheim, it was designed to treat inflammatory conditions by inhibiting the LTB4-mediated recruitment and activation of neutrophils. Amelubant is a prodrug that is rapidly metabolized into its active form, BIIL 260, upon administration. It was primarily investigated for the treatment of chronic obstructive pulmonary disease (COPD), asthma, cystic fibrosis, and rheumatoid arthritis. However, clinical development was largely halted after Phase II trials failed to demonstrate sufficient therapeutic benefit over existing treatments or placebo in these indications.
