Overview

Amenamevir is a small molecule antiviral drug that acts as a helicase-primase inhibitor. It is primarily used for the treatment of herpes zoster (shingles) and has demonstrated activity against varicella-zoster virus (VZV) and herpes simplex viruses types 1 and 2 (HSV-1, HSV-2). Unlike nucleoside analogs such as acyclovir, amenamevir inhibits viral DNA replication by targeting the helicase–primase complex essential for viral DNA synthesis. This mechanism allows it to be effective even in cells with abundant viral DNA synthesis or high viral load. Amenamevir was discovered by Astellas Pharma and was approved in Japan in 2017 for the treatment of herpes zoster[1][5][6][7].
