Drug intelligence / Profile preview
AMEV2 (Acinetobacter Multi-Epitope Vaccine 2) is an immunoinformatics-designed recombinant multi-epitope subunit vaccine developed to prevent multidrug-resistant (MDR) *Acinetobacter baumannii* infections. The vaccine construct consists of an *A. baumannii* thioredoxin A (TrxA) leading protein sequence followed by five tandemly linked peptide antigens containing both putative B- and T-cell epitopes. These five peptide antigens are derived from key proteins associated with *A. baumannii* pathogenesis: copper homeostasis protein NlpE, outer membrane nuclease NucAB, siderophore receptor TonB, zinc piracy receptor ZnuD, and outer membrane protein Omp38. Subcutaneous immunization with AMEV2, typically formulated with the AddaS03 adjuvant system, elicits robust humoral and cellular immune responses, including high titers of antigen-specific antibodies and increased frequencies of antibody-secreting cells. The protective mechanism of AMEV2 is primarily antibody-mediated, acting through complement- and Fc gamma receptor-mediated opsonophagocytic killing of *A. baumannii* by macrophages.
