Overview

AMG 172 is an antibody-drug conjugate (ADC) developed by Amgen for the treatment of cancer, specifically targeting relapsed or refractory clear cell renal cell carcinoma (ccRCC). It consists of a human IgG1 monoclonal antibody directed against CD27L (also known as CD70 antigen), conjugated via a non-cleavable linker to the cytotoxic agent maytansinoid DM1. The mechanism of action involves binding to CD27L on tumor cells, internalization of the ADC, and subsequent release of DM1 inside the cell, leading to microtubule inhibition and cell death. Clinical studies have shown that AMG 172 has a favorable pharmacokinetic profile but limited antitumor activity in ccRCC patients[1][2][3][7].
