AMG-222

AMG-222 is an orally active, small molecule inhibitor of dipeptidyl peptidase IV (DPP-IV, also known as CD26 antigen). DPP-IV inactivates glucagon-like peptide 1 (GLP‑1), a key mediator of blood glucose regulation after meals. By inhibiting DPP-IV, AMG‑222 increases endogenous GLP‑1 levels, which can improve glycemic control and enhance pancreatic beta cell function. This mechanism represents a novel approach to the treatment of type 2 diabetes mellitus. The drug was originally developed by Alantos Pharmaceuticals and later acquired by Amgen for further development[1][4]. Clinical development reached phase II trials for type 2 diabetes but was subsequently discontinued[4].
