Overview

AMG 228 is an agonistic human IgG1 monoclonal antibody that targets the glucocorticoid-induced tumor necrosis factor receptor-related protein (GITR), also known as CD357, which is expressed on effector and regulatory T cells. By binding to and activating GITR, AMG 228 stimulates the immune system by promoting activation and proliferation of tumor-antigen-specific T-effector cells while suppressing the function of activated T-regulatory cells. This dual action aims to enhance anti-tumor immune responses. Developed by Amgen, AMG 228 was investigated primarily for the treatment of advanced solid tumors in a first-in-human phase 1 clinical trial. The drug was administered intravenously every three weeks and demonstrated acceptable safety with no dose-limiting toxicities up to the highest tested dose; however, there was no evidence of significant anti-tumor activity or robust T-cell activation in patients[1][3][4][5][6][7].
