Overview

AMG 317 is a fully human monoclonal antibody that targets the interleukin-4 receptor subunit alpha (IL-4Rα), thereby blocking both IL-4 and IL-13 signaling pathways. These cytokines are key mediators in allergic airway inflammation and play a significant role in the pathogenesis of asthma. Developed by Amgen as an antiasthmatic agent, AMG 317 was evaluated for efficacy and safety in patients with moderate to severe asthma. While it demonstrated some clinical activity—particularly among patients with more severe or uncontrolled symptoms—it did not show statistically significant efficacy across the overall study population. The drug was well tolerated but its development for asthma has been discontinued[1][2][3][5].
