AMG-337 is an orally bioavailable, highly selective small molecule inhibitor of the proto-oncogene c-Met (also known as MET receptor tyrosine kinase). It acts as an ATP-competitive inhibitor, binding to and blocking the activity of c-Met, thereby disrupting downstream signaling pathways involved in cell proliferation, survival, invasion, and morphogenesis. Dysregulation or amplification of c-Met is implicated in various cancers. AMG-337 has been investigated primarily for the treatment of advanced solid tumors—especially those with MET amplification—including gastric cancer, esophageal cancer, sarcoma, and other solid tumors. The drug was developed by Amgen and later also by NantPharma[1][3][4][5][6].
