AMG 340 is a bispecific antibody designed as a T-cell engager (TCE) that targets both prostate-specific membrane antigen (PSMA) on prostate cancer cells and the CD3 receptor on T cells. It features a novel low-affinity anti-CD3 binding domain, specifically engineered to reduce the risk and severity of cytokine release syndrome (CRS), which is a common toxicity associated with this class of immunotherapies. The drug was developed for use in metastatic castration-resistant prostate cancer (mCRPC). In clinical trials, AMG 340 demonstrated manageable safety with step-up dosing but did not show sufficient clinical activity to warrant further development. The drug was originally developed by TeneoBio and later by Amgen[1][2][6][7].
