Overview

AMG 655 + paclitaxel + carboplatin is a combination regimen used experimentally in oncology, where **AMG 655 (conatumumab)** is a fully human IgG1 agonist monoclonal antibody that targets **death receptor 5 (DR5)**, activating apoptotic pathways in tumor cells through caspase activation. Paclitaxel is a microtubule stabilizer, inducing cell cycle arrest, and carboplatin is a platinum-based agent that crosslinks DNA and induces apoptosis. AMG 655 amplifies the apoptotic effect of cytotoxic agents like paclitaxel and carboplatin by potentiating death receptor signaling. The combination is designed to enhance antitumor activity, primarily in solid tumors where apoptosis resistance may limit chemotherapy efficacy. Amgen developed AMG 655 as a targeted immunotherapy.
