Overview

AMG 780 is a fully human IgG2 monoclonal antibody developed to target the proangiogenic cytokines angiopoietin 1 (Ang1) and angiopoietin 2 (Ang2). By binding to Ang1 and Ang2, AMG 780 inhibits their interaction with the Tie2 receptor, thereby blocking angiogenesis—a process critical for tumor growth and metastasis. The drug was investigated primarily for its potential anti-angiogenic and antineoplastic activities in patients with advanced solid tumors. Clinical studies demonstrated that AMG 780 could be administered intravenously at doses up to 30 mg/kg every two weeks, showing dose-proportional pharmacokinetics and manageable toxicity consistent with other antiangiopoietin therapies. However, no objective tumor responses were observed in early-phase trials, though some patients achieved stable disease[1][2][3][4].
