AMG 811 is a fully human IgG1 monoclonal antibody developed by Amgen that selectively targets and neutralizes human interferon gamma (IFN-γ). Its mechanism of action involves binding to IFN-γ and inhibiting its activity, thereby modulating downstream immune responses associated with autoimmune diseases. AMG 811 was investigated primarily for the treatment of systemic lupus erythematosus (SLE), including lupus nephritis, discoid lupus erythematosus (DLE), and psoriasis. Clinical studies demonstrated that while AMG 811 effectively reduced IFN-γ-associated biomarkers such as CXCL10 protein levels and gene expression signatures in blood and skin, it did not show significant clinical efficacy in improving disease outcomes. The drug exhibited an acceptable safety profile but development has been discontinued for these indications[1][2][3][5][7].
