AMG 820

AMG 820 is a fully human IgG2 monoclonal antibody that targets the colony-stimulating factor 1 receptor (CSF1R, also known as c-fms). By binding to CSF1R, AMG 820 blocks the interaction of its ligands—colony-stimulating factor 1 (CSF-1) and interleukin-34 (IL-34)—with the receptor. This inhibition suppresses CSF-1-induced signaling pathways, leading to reduced recruitment and activation of tumor-associated macrophages (TAMs) in the tumor microenvironment. TAMs are known to promote an immunosuppressive environment that supports tumor growth. The drug was developed for use in advanced solid tumors, including colorectal cancer and other malignancies resistant or refractory to PD-1 inhibitors. Clinical studies have shown pharmacodynamic effects but limited antitumor activity when used alone or in combination with pembrolizumab[1][2][3][6].
