Drug intelligence / Profile preview
**AMG504-1** is a needle-free, intranasal dry powder glucagon formulation developed for the treatment of severe hypoglycemia, particularly in patients with diabetes who may struggle with injectable rescue therapies. It consists of 2 mg or 3 mg glucagon in a compact, single-use nasal powder dosing device for simple one-step administration into one nostril. Glucagon activates hepatic receptors to stimulate glycogenolysis and gluconeogenesis, rapidly increasing blood glucose levels; clinical studies in healthy volunteers, adults, and children with type 1 diabetes demonstrated efficacy comparable to intramuscular glucagon, with a \u226525 mg/dL glucose rise within 20 minutes in nearly all cases, alongside a favorable safety profile featuring transient mild nasal irritation but reduced nausea versus injections. Originally developed by Locemia Solutions (previously A.M.G Medical Inc.), it was investigated in Phase 1 pediatric trials and later advanced by partners like Eli Lilly (as LY900018) and Amphastar Pharmaceuticals, paving the way for approved intranasal glucagon products like Baqsimi.[1][4][7][17]
