Overview

Amifostine liposomes are a novel nanoparticle formulation of the cytoprotective agent amifostine, designed to enhance radiation protection while minimizing the systemic toxicity associated with the small-molecule drug. This formulation consists of pegylated liposomes that encapsulate or are coated with amifostine. In preclinical studies, these liposomes have demonstrated the ability to scavenge reactive oxygen species (ROS), reduce DNA damage, and protect mitochondria from radiation-induced injury. The mechanism of action involves the regulation of glutathione peroxidase 4 (GPX4) and the inhibition of ferroptosis (iron-dependent cell death). By altering the pharmacokinetic profile—primarily distributing to the liver and kidney—this liposomal delivery system aims to provide a more effective and safer alternative for mitigating radiation-induced tissue damage and systemic side effects.
