Drug intelligence / Profile preview
The amikacin + fosfomycin inhalation solution is a fixed-dose combination of two antibiotics—amikacin (an aminoglycoside) and fosfomycin (a phosphonic acid derivative)—formulated for aerosol delivery via an in-line nebulizer system. This investigational therapy is designed as adjunctive treatment for pneumonia caused by Gram-negative bacteria in mechanically ventilated patients. The combination exploits the synergistic antibacterial effects of both drugs to enhance efficacy against multidrug-resistant Gram-negative pathogens and also provides coverage against some Gram-positive organisms such as methicillin-resistant Staphylococcus aureus (MRSA). Amikacin acts by binding to the 30S ribosomal subunit, inhibiting bacterial protein synthesis, while fosfomycin inhibits bacterial cell wall synthesis by blocking the enzyme MurA. The drug is delivered using a specialized nebulizer system (eFlow Inline), which enables efficient deposition of high concentrations directly into the lungs with minimal systemic exposure[1][5][6][7].
