Overview

Aminaphthone is a synthetic small molecule derived from 4-aminobenzoic acid, used for more than thirty years in the treatment of various vascular disorders, especially chronic venous insufficiency and capillary fragility[1][2][4]. It acts as an endothelial protector by stabilizing endothelial cells and reducing capillary permeability. Aminaphthone modulates the expression of VE-cadherin and downregulates adhesion molecules (VCAM, ICAM, selectins), vasoconstrictor peptides (endothelin‑1), and pro-inflammatory cytokines (IL‑6, IL‑10, VEGF, TGF-beta)[1][4][8]. Its mechanism supports improved microvascular reactivity in conditions such as Raynaud’s phenomenon and systemic sclerosis[4][8]. The drug is partially metabolized to phthiocol and excreted via urine[8].
