Overview

Aminoglutethimide + danazol + tamoxifen is a combination of three small molecule drugs used as a hormonal therapy regimen, primarily studied in postmenopausal women with metastatic breast cancer. Each component acts via distinct mechanisms: - Aminoglutethimide is an adrenocortical steroid synthesis inhibitor that blocks the conversion of cholesterol to pregnenolone and inhibits aromatase, reducing estrogen production. - Danazol is a synthetic androgen and gonadotropin inhibitor that suppresses pituitary output of follicle-stimulating hormone (FSH) and luteinizing hormone (LH), leading to decreased estrogen synthesis. - Tamoxifen is a selective estrogen receptor modulator (SERM) that competitively inhibits estrogen binding to its receptor, blocking the proliferative action of estrogens on breast tissue. This combination was investigated for synergistic effects in suppressing tumor growth in hormone-sensitive metastatic breast cancer. However, subsequent studies suggested potential antagonism between aminoglutethimide and danazol regarding estradiol bioavailability[1][2][3][4].
