Overview

Aminoglycosides + β-lactams is a combination of two classes of antibiotics frequently used together for the treatment of severe bacterial infections, particularly those caused by Gram-negative organisms. Aminoglycosides (such as gentamicin, tobramycin, or amikacin) act primarily by binding to the 30S ribosomal subunit and inhibiting bacterial protein synthesis, leading to bactericidal activity. They also disrupt the outer membrane of bacteria, increasing permeability. β-Lactams (such as penicillins or cephalosporins) inhibit bacterial cell wall synthesis by binding to penicillin-binding proteins (PBPs), resulting in cell lysis and death. The combination is often synergistic because β-lactams can increase aminoglycoside uptake into bacteria by disrupting the cell wall. This regimen is commonly used for serious infections like endocarditis and sepsis due to Enterococcus species or other resistant Gram-negative pathogens[1][3][7].
