Overview

Aminophenazone is a small molecule analgesic and antipyretic of the pyrazolone class, historically used for pain relief and fever reduction in conditions such as rheumatism, neuritis, pulmonary tuberculosis, and common colds. It was also used in combination with ergotamine and caffeine for acute migraine attacks. Aminophenazone acts as an anti-inflammatory agent but carries a significant risk of life-threatening agranulocytosis, which led to its withdrawal from many markets. The drug has also been utilized in radiolabeled form (13C-labeled) for non-invasive liver function tests measuring cytochrome P450 metabolic activity. Its mechanism involves inhibition of prostaglandin synthesis via cyclooxygenase inhibition[1][2][3][4][5][7].
