Overview

Amitifadine is an orally administered small molecule classified as a "triple reuptake inhibitor" that inhibits the reuptake of serotonin, norepinephrine, and dopamine. This mechanism increases extracellular concentrations of these three monoamines in the brain. Amitifadine was developed primarily as an antidepressant and has demonstrated statistically significant antidepressant effects in clinical trials for major depressive disorder. It has also been investigated for other indications such as obesity, substance use disorders (including nicotine and opioid addiction), pain, alcoholism, and smoking withdrawal. The drug was originally developed by DOV Pharmaceutical and later by Euthymics Bioscience and Ethismos Research[3][5][6]. Its development for major depressive disorder and obesity was discontinued after reaching Phase 3 clinical trials[3][6].
