Overview
Amiclo is a fixed-dose combination drug developed by Popular Pharmaceuticals, containing the tricyclic antidepressant amitriptyline and the benzodiazepine anxiolytic chlordiazepoxide. It is indicated for the treatment of patients with moderate to severe anxiety associated with depression. Amitriptyline acts primarily by inhibiting the reuptake of norepinephrine and serotonin at the presynaptic neuronal membrane, thereby increasing their concentration in the synaptic cleft. Chlordiazepoxide enhances the inhibitory effect of gamma-aminobutyric acid (GABA) by binding to the benzodiazepine site on the GABA-A receptor complex, leading to anxiolytic, sedative, and muscle-relaxant effects. This combination is typically used when both depressive and anxiety symptoms are prominent.
