Overview

Combination of three pharmaceuticals used or being studied for the treatment of certain cancers, particularly non-small cell lung cancer (NSCLC) and head and neck squamous cell carcinoma. - **Amivantamab** is a fully human bispecific monoclonal antibody targeting both the Epidermal Growth Factor Receptor (EGFR) and the MET receptor, leading to receptor degradation and inhibition of downstream signaling, suppressing tumor growth and progression. - **Pembrolizumab** is a monoclonal antibody, specifically an immune checkpoint inhibitor targeting PD-1, which enhances antitumor immune response by preventing the inactivation of T cells. - **Carboplatin** is a platinum-based chemotherapeutic, which forms DNA crosslinks leading to apoptosis of rapidly proliferating cells. The combination of these agents is under clinical development for solid malignancies, with primary investigation in NSCLC and recurrent/metastatic head and neck cancer. In NSCLC, the closely related combination of amivantamab + carboplatin + pemetrexed has marketing approval, but the addition of pembrolizumab is under clinical investigation.
