Overview

Amivantamab hyaluronidase is a subcutaneous formulation of amivantamab, a fully human bispecific antibody targeting the epidermal growth factor receptor (EGFR) and mesenchymal-epithelial transition (MET) receptor. It is co-formulated with recombinant human hyaluronidase (rHuPH20) using Halozyme's ENHANZE technology, which temporarily degrades hyaluronan in the subcutaneous space to facilitate the administration of large volumes. The mechanism of action involves blocking ligand binding to EGFR and MET, promoting receptor internalization and degradation, and inducing antibody-dependent cellular cytotoxicity (ADCC) and antibody-dependent cellular phagocytosis (ADCP). Developed by Janssen (Johnson & Johnson), this formulation is designed to reduce administration time and infusion-related reactions compared to the intravenous version. It is primarily indicated for patients with non-small cell lung cancer (NSCLC) harboring specific genetic alterations, including EGFR exon 20 insertion mutations and MET amplification.
