Amivutamab (SL-101) is a fully human monoclonal antibody that targets the epidermal growth factor receptor (EGFR). Developed by Shanghai Sailon Biotechnology, it is specifically being evaluated for the treatment of patients with RAS wild-type metastatic colorectal cancer (mCRC) who have progressed after standard therapies. The antibody binds to the extracellular domain of EGFR, preventing the binding of ligands such as epidermal growth factor (EGF) and transforming growth factor-alpha (TGF-α). This blockade inhibits the activation of intracellular signaling cascades, including the MAPK and PI3K/Akt pathways, which are critical for tumor cell proliferation, survival, and angiogenesis. As a fully human antibody, amivutamab is designed to offer a favorable safety profile with potentially lower immunogenicity compared to chimeric anti-EGFR agents like cetuximab.
