**AML-VAC-XS15** is an investigational, off-the-shelf multipeptide therapeutic cancer vaccine for prevention of relapse in adults with acute myeloid leukemia in complete remission or complete remission with incomplete blood-count recovery after first-line therapy. It contains nine immunopeptidomics-defined AML and leukemia progenitor-cell antigen peptides: two HLA class I-restricted neoepitopes derived from mutant NPM1, one HLA class II-restricted neoepitope derived from IDH2 R140Q, and six non-mutated, HLA class II-restricted AML and leukemia progenitor-cell-associated peptides. The peptide mixture is adjuvanted with XS15, a synthetic Toll-like receptor 1/2 agonist, and emulsified with Montanide ISA 51 VG for subcutaneous administration. The vaccine is intended to generate durable antigen-specific CD4-positive and CD8-positive T-cell responses against residual AML cells, including relapse-initiating leukemia progenitor and stem cells.
