This drug is a fixed-dose triple combination therapy developed by Zentiva (under its Zent2U B2B brand) for the treatment of hypertension. It combines three established antihypertensive agents with complementary mechanisms of action: amlodipine besilate, a dihydropyridine calcium channel blocker (CCB); candesartan cilexetil, an angiotensin II receptor blocker (ARB); and indapamide, a thiazide-like diuretic. Amlodipine induces vasodilation by inhibiting calcium influx into vascular smooth muscle cells; candesartan blocks the vasoconstrictive and aldosterone-secreting effects of angiotensin II at the AT1 receptor; and indapamide promotes sodium and water excretion by inhibiting the sodium-chloride symporter in the distal convoluted tubule. This triple combination is designed to provide robust blood pressure control in patients who are not adequately managed on dual therapy, simplifying treatment regimens and potentially improving adherence.
