AD-209-1B is a dual fixed-dose combination (FDC) of amlodipine and chlorthalidone, developed by Addpharma Inc. for the treatment of essential hypertension. Amlodipine is a dihydropyridine calcium channel blocker (CCB) that inhibits the influx of calcium ions into vascular smooth muscle and cardiac muscle, resulting in peripheral vasodilation and reduced blood pressure. Chlorthalidone is a long-acting thiazide-like diuretic that inhibits sodium and chloride reabsorption in the distal convoluted tubule of the nephron by targeting the sodium-chloride cotransporter (NCC), leading to increased excretion of sodium and water and a subsequent reduction in extracellular fluid and plasma volume. This specific formulation, containing 2.5 mg of amlodipine and 6.25 mg of chlorthalidone, is utilized as a comparator arm in clinical development programs for the triple combination therapy AD-209 (amlodipine/chlorthalidone/telmisartan).
